چکیده

To date, the human G-protein beta 3 subunit (GNB3) gene and some of its variants represent some of the best examples of genetic influences that are involved in the determination of hypertension and obesity, which make it a sensible candidate gene for type 2 diabetes. To assess the influence of GNB3 in type II diabetes mellitus (NIDDM), we carried out a retrospective, case-control study of variant GNB3 825C>T for putative correlations with NIDDM amongst nationals from the United Arab Emirates (Emirati) - an ethnic group characterized by no alcohol intake and no cigarette smoking. We investigated a sample population of 510 Emirati (257 men, 253 women) comprising two groups - 254 controls and 256 patients with clinical diagnoses of type 2 diabetes (cases). The GNB3 C825T dimorphism showed an association with NIDDM Chi2 =22.5, 2 df, P<0.001). Further analysis revealed that the GNB3 T/T 825 genotype was positively associated with NIDDM (Yates corrected Chi2=20.6, 2 df, P<0.001; odds ratio of 2.44 with a 95% confidence interval of 1.64 - 3.63) compared to pooled CC/CT genotypes. Our data shows that GNB3 T825 allele may be involved in the pathogenesis of DM through a pathway that is different from the one implicated in obesity.

The Association between GNB3 Gene Polymorphism and Obesity: A Systematic Review and Meta-Analysis

The association between Guanine Nucleotide Binding protein β protein polypeptide 3(GNB3) C825T polymorphism and obesity has recently been reported. However, the findings remain inconclusive. The aim of this systematic review and meta-analysis was to detect the relationship between GNB3 C825T polymorphism and obesity. Materials and Methods: Six electronic databases including Embase, Medlin...

G protein beta 3 gene: structure, promoter, and additional polymorphisms.

Recent studies have shown that a polymorphism (C825T) in the gene encoding the G protein beta 3 subunit (GNB3) is associated with hypertension and obesity. We characterized the entire GNB3 gene, which spans 7.5 kb and is composed of 11 exons and 10 introns. Its promoter lacks a TATA box but harbors GC-rich regions. The functional activity of the GNB3 promoter was verified with reporter gene ass...

G protein beta3 subunit variant and essential hypertension in Japanese.

Enhanced G protein activation has been implicated to underlie the increased sodium-proton transport in blood cells, an impaired characteristic observed in 30% to 50% of patients with essential hypertension. Recently, significant association between a C825T polymorphism of the gene encoding the G protein beta3 subunit and hypertension was demonstrated in a white population, together with the fin...

The human G protein beta 3 subunit GNB3 825T polymorphism.

Meirhaeghe et al. recently showed that the T allele of a C vs T exchange at position 825 of the gene encoding for the beta3 subunit of heterotrimeric G proteins (GNB3) is associated with enhanced coronary constriction after intracoronary administration of methylergonovine. The 825T allele has also been shown to be associated with an enhanced response after activation of G protein coupled recept...

GNB3 overexpression causes obesity and metabolic syndrome

The G-protein beta subunit 3 (GNB3) gene has been implicated in obesity risk; however, the molecular mechanism of GNB3-related disease is unknown. GNB3 duplication is responsible for a syndromic form of childhood obesity, and an activating DNA sequence variant (C825T) in GNB3 is also associated with obesity. To test the hypothesis that GNB3 overexpression causes obesity, we created bacterial ar...
